Question: can groups only be defined in the Main web ($TWiki::cfg{UsersWebName} = 'Main'). I think that this is reasonable to presume as searching for groups will be speeded up search for them only in the Main web.

it certainly is sped up on larger twikis running on comparitively slow disks. And while i'd rather that users and groups could be in all sorts of webs, it certainly makes sense to restrict groups in the same way as user topics are..
